Spira Grujić was born on December 7, 1971, in Pristina (then part of Yugoslavia). He began his career with Red Star Belgrade in the youth ranks before settling at Radnički Niš, where he played from 1991 to 1995, making 58 appearances and scoring three goals. In 1995, he moved to Belgium, playing for Molenbeek for two seasons and then for Anderlecht in the 1997–98 season, before moving to the Netherlands in 1998 to play for FCTwente, after Nico-Jan Hoogma departure to Hamburger SV.

At Twente, between 1998 and 2004, he became the leader and the sheriff of the Tukkers' defense. He played a total of 169 matches and scored three goals, being part of the starting lineup that won the KNVB Beker in 2001. In his final years with the club, he served as captain and became a beloved figure among the fans: fans would often chant "Spi-ra, Spi-ra" in support of the player inside the stadium.

His consistency was evident in every match: he was a strong tackler, with excellent reading of the game and positioning, and a strong work ethic on the field. Measuring 1,85m, Grujic was physically imposing, further complementing his character as a defender with a combative mentality, but also possessed a good passing ability to initiate attacking build-ups. He was frequently praised for his winning mentality and commitment to the team.

After leaving FC Twente in 2004, he spent two more seasons at ADO Den Haag, where he successfully helped the Hague side escape relegation twice, before ending his career at FK Rad, his club in Serbia, in 2007. At international level, Grujić made only one appearance for the Yugoslavia senior team, appearing in a friendly against Northern Ireland on 16 August 2000, without scoring any goals.
